Optical clearing alters the sensitivity of laser speckle contrast imaging to flow velocity

Abstract
Optical clearing improves visibility but can bias laser speckle contrast imaging (LSCI). We combined OCT and PCA-LSCI in separate paired agarose phantoms during fluorescein diffusion. Speckle metrics changed at constant flow, and DAC sensitivity to velocity increased with clearing. Local background correction increased the velocity-associated share of variance from 39.8% to 56.3% and reduced the concentration-associated share from 41.2% to 15.4%. Quantitative flow monitoring therefore requires calibration for the local optical state.
