Microcirculatory Response of Tumor and Healthy Tissues to Various Photodynamic Therapy Protocols

Abstract
Photodynamic therapy is an effective method for oncological diseases treatment; however, the ongoing refinement of its protocols necessitates the development of objective methods to assess treatment outcomes. The state of the microcirculatory bed is one of the most informative indicators. This study presents a non-invasive method and setup for the quantitative assessment of skin microcirculation using videocapillaroscopy and photoplethysmography, enabling direct monitoring during photoactivation. The system was tested in a study of the microcirculatory response in intact rats and in animals with a cholangioma model. Measurements were performed in the tumor area and surrounding tissue for two irradiation modes: continuous and pulsed. Parameters for evaluating dynamic changes in the density of active vessels and tissue blood perfusion were introduced. The difference in the vascular response between tumor and healthy tissues was quantitatively characterized, and it was established that this response also depends on the treatment protocol. The development of the proposed approach may be valuable for assessing the efficacy of photodynamic therapy when optimizing its protocols.
