Engineering Strategies to Overcome Biological Barriers

Abstract

The clinical success of modern biopharmaceuticals is frequently hindered by rapid systemic clearance, poor site-specificity, and unfavorable immunogenic responses. Overcoming these hurdles requires a paradigm shift from simple drug formulation to the design of sophisticated delivery systems capable of navigating the complex biological landscape. This presentation explores the current landscape of drug delivery, focusing on the development of smart, bio-inspired, and stimuli-responsive nanocarriers.

The transition towards next-generation delivery platforms will be discussed, highlighting the unique role of ferritin-based nanocages as robust, protein-derived vehicles that offer high loading capacity, inherent biocompatibility, and ease of surface functionalization for precise targeting. Furthermore, we will examine the development of pH-sensitive delivery systems designed to exploit the tumor microenvironment and endolysosomal pathways, ensuring efficient intracellular release while minimizing systemic toxicity.

The talk will also address the critical challenge of immunogenicity and stability, evaluating how advanced encapsulation strategies can modulate the interaction between the carrier and the host immune system. By integrating material science with molecular biology, we demonstrate how these intelligent architectures move beyond mere transport to provide controlled, safe, and effective therapeutic outcomes. Finally, we will outline current technical challenges and future perspectives in transitioning these systems from in vitro optimization to clinical application.
